{"id":1507,"date":"2022-08-07T18:15:23","date_gmt":"2022-08-07T16:15:23","guid":{"rendered":"https:\/\/solmed-clinic.com\/?p=1507"},"modified":"2022-08-09T14:43:54","modified_gmt":"2022-08-09T12:43:54","slug":"targeted-blockade-treatment-crohnove-bolesti","status":"publish","type":"post","link":"https:\/\/solmed-clinic.com\/en\/targeted-blockade-treatment-crohnove-bolesti\/","title":{"rendered":"Targeted blockade of interleukin-23 &#8211; is this how we treat the cause of Crohn&#8217;s disease?"},"content":{"rendered":"<p><span style=\"font-weight: 400;\">To date, a strong link has been established between the polymorphism of the interleukin-23 (IL -23) mediator gene and the risk for the development, treatment, and outcome of various autoimmune diseases, but most of the evidence on this topic has been gathered in Crohn&#8217;s disease patients.\u00a0<\/span><\/p>\n<h2><span style=\"font-weight: 400;\">What is it really about?\u00a0<\/span><\/h2>\n<p><span style=\"font-weight: 400;\">Receptors for IL -23 on T lymphocytes bind IL -23 molecules twice as strongly, meaning that a normal concentration of IL -23 in the blood can overactive T lymphocytes, which activate another cascade that causes Crohn&#8217;s disease. Blocking this receptor may prevent another cascade that causes Crohn&#8217;s disease flare-ups.\u00a0<\/span><\/p>\n<h3><span style=\"font-weight: 400;\">There have been three groups of drugs approved for the treatment of Crohn&#8217;s disease:\u00a0<\/span><\/h3>\n<ul>\n<li><span style=\"font-weight: 400;\">TNF-\u03b1 antagonists, which reverse the harmful effects of TNF-\u03b1 molecules<\/span><\/li>\n<li><span style=\"font-weight: 400;\">integrin inhibitors, which prevent all leukocytes from entering the tissues and thus also &#8220;put out the fire&#8221;<\/span><\/li>\n<li><span style=\"font-weight: 400;\">IL -antagonists. -12 and IL -23, which alone potentially act at the beginning of this inflammatory cascade.\u00a0<\/span><\/li>\n<\/ul>\n<p><span style=\"font-weight: 400;\">The realization that the sensitive receptor for IL -23 is indeed the main culprit in Crohn&#8217;s disease has also led to the discovery of drugs that specifically block IL -23 receptors.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Indeed, in the early stages of the study, this group of drugs proved to be effective &#8220;joint&#8221; inhibitors of IL -12 and IL -23 in Crohn&#8217;s disease treatment, psoriasis, and psoriatic arthritis.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Now it is time for a great Phase 3 study to compare the efficacy of a selective IL -23 blocker and a dual IL -23 and IL -12 blocker.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">We are pleased that Solmed and Dr. Dominik Kralj are involved in this great project.<\/span><\/p>\n<p><a href=\"https:\/\/docs.google.com\/forms\/d\/e\/1FAIpQLSeSLczrxAhDG0c9lDy63t2lhlIJRXSAoVNUq419NSn0s3bysA\/viewform?usp=sf_link\"><span style=\"font-weight: 400;\"><span style=\"text-decoration: underline;\">Click Here<\/span><\/span><\/a><span style=\"font-weight: 400;\"> to fill out our questionnaire and see if you meet the criteria to receive free screening and treatment!<\/span><\/p>\n","protected":false},"excerpt":{"rendered":"<p>To date, a strong link has been established between the polymorphism of the interleukin-23 (IL -23) mediator gene and the risk for the development, treatment, and outcome of various autoimmune &#8230;<\/p>\n","protected":false},"author":4,"featured_media":1508,"comment_status":"closed","ping_status":"closed","sticky":false,"template":"","format":"standard","meta":{"_acf_changed":false,"inline_featured_image":false,"footnotes":""},"categories":[8,38,48],"tags":[72,78],"class_list":["post-1507","post","type-post","status-publish","format-standard","has-post-thumbnail","hentry","category-blog-en","category-clinical-research","category-treatment","tag-crohns-disease","tag-gastroentology"],"acf":[],"yoast_head":"<!-- This site is optimized with the Yoast SEO plugin v27.1.1 - https:\/\/yoast.com\/product\/yoast-seo-wordpress\/ -->\n<title>Targeted blockade of interleukin-23 - is this how we treat the cause of Crohn&#039;s disease?
